Dean felt a great sense of relief when the mark of Cain was finally removed from him. In theory, it was supposed to take away all that was associated with it, including the undeniable attraction he had for Amara. But something went wrong. While old flames like Jo and Lisa have begun to surface in his life, he still has this bond to Amara that he can’t shake.
Sam Winchester

In attempts to destroy Amara, the darkness, the mark of Cain was to be transferred from her to Sam. However, it was thought that this process was reversed. Unbeknownst to anyone but Sammy, a mark was left on him, so pale that it is barely noticeable. Sam can sure as hell feel it though, burning on him quite painfully. He now has begun to understand the dark feelings Dean once had when he bore the mark. Only this time, he’s getting the longing for demon blood once more. The worst part of it all is that it’s been rumored Ruby’s back in town, perhaps to feed into his former addiction.
John Winchester

Mary is pretty open to interpretation based loosely on the few blurbs we’ve seen of her. Maybe she has a cool[or terrifying] story about what happened to her in the afterlife. Whether or not she is just a mortal or a hunter when she returns to life is completely up to you. However, her most important goal should be to reunite with her family with her second chance at life.
Adam Milligan

The last we heard of Adam, he was still stuck in hell after Sam and Dean failed[or frankly just forgot] to rescue him. You can make him whatever you’d like. It’d be pretty cool, for drama’s sake, if he traded his soul or whatever for a set of black eyes. In having the power of a demon, he can find his revenge on his brothers for abandoning him. Or, if you just want him to come back as a mortal and try to join in on their hunting gang, that’s fine too. Another possibility is to have him completely unaware of Sam/Dean[seeing as he was actually eaten by a ghoul, then used as a vessel for Michael] and play out there completely confusion in seeing him alive.
Castiel

Might be interesting if he has some damage or residual effects from Lucifer having used him as a vessel but totally up to you. Lots of awkward questions and please make him say assbutt a couple times. hehe.

Crowley and Lucifer are now duking it out over who is king of hell. Both reside in hell and refuse to move, fighting on a constant basis. As hell more or less exists on earth, they have brought things to the surface and of course, are acting as immature as ever.

Not too long ago, it was revealed that Chuck is actually god, disguising himself after all this time as a prophet of the Lord. Chuck has a lot of stuff to work through, particularly with Amara. The two went away on hiatus to try to repair their relationship. He is still weak from her attempt to initially defeat him and for that reason, won’t be much help physically at the start.

When Amara and Chuck left to reconcile their sibling relationship, something with their magic put a strain on the earth. A portal was broken and it is believed to have been an accident. It’s up to you, but perhaps this was Amara’s intention and her act of making up with her brother is simply a guise.
